RESUMO

Colorectal cancer is a malignant tumour with high incidence and mortality worldwide; therefore, improving the early diagnosis of colorectal cancer and implementing a targeted "individualized treatment" strategy is of great concern. NIR-II fluorescence imaging is a large-depth, high-resolution optical bioimaging tool. Around the NIR-II window, researchers have developed a variety of luminescent probes, imaging systems, and treatment methods with colorectal cancer targeting capabilities, which can be visualized and image-guided in clinical surgery. This article aims to overcome the difficulties in diagnosing and treating colorectal cancer. The present review summarizes the latest results on using NIR-II fluorescence for targeted colorectal cancer imaging, expounds on the application prospects of NIR-II optical imaging for colorectal cancer, and discusses the imaging-guided multifunctional diagnosis and treatment platforms.

Lactobacillus strains isolated from female athletes were tested. MRS broth was supplemented with a range of Metronidazole concentrations from 128 to 2000 μg/ml. The growth of these strains was monitored by measuring optical density at 3-hour intervals over 24 hours. Results: It was observed that at Metronidazole concentrations up to 128 μg/ml, there was no significant impact on the growth of the Lactobacillus strains. However, at concentrations above 512 μg/ml, Metronidazole significantly inhibited their growth. The response to Metronidazole varied among different Lactobacillus strains. For instance, at a concentration of 256 μg/ml, Lactobacillus delbrueckii, Lactobacillus jensenii, and Lactobacillus vaginalis showed notable inhibition, whereas Lactobacillus crispatus, Lactobacillus gasseri, and Lactobacillus fermentum were not significantly affected. Discussion: High concentrations of Metronidazole were found to inhibit the growth of the six Lactobacillus strains isolated from female athletes. Lower concentrations had negligible effects. The differential response of Lactobacillus strains to varying concentrations of Metronidazole (between 128 μg/ml and 512 μg/ml) highlights the need for careful consideration of Metronidazole use in managing vaginal microbiota health in female athletes (AU)

Importance: Epidural anesthesia is a primary choice for cesarean delivery, but supplemental analgesics are often required to relieve pain during uterine traction. Objective: To investigate the sedative and analgesic effects of intravenous esketamine administered before childbirth via cesarean delivery with the patient under epidural anesthesia. Design, Setting, and Participants: This multicenter, double-blind randomized clinical trial assessed 903 women 18 years or older who had full-term single pregnancy and were scheduled for elective cesarean delivery with epidural anesthesia in 5 medical centers in China from September 18, 2021, to September 20, 2022. Intervention: Patients were randomized to receive intravenous injection of 0.25 mg/kg of esketamine or placebo before incision. Main Outcomes and Measures: The coprimary outcomes included scores on the numeric rating scale of pain (an 11-point scale, with 0 indicating no pain and 10 indicating the worst pain; a difference of ≥1.65 points was clinically meaningful) and Ramsay Sedation Scale (a 6-point scale, with 1 indicating restlessness and 6 indicating deep sleep without response; a difference of ≥2 points was clinically meaningful) immediately after fetal delivery. Secondary outcomes included neonatal Apgar score assessed at 1 and 5 minutes after birth. Results: A total of 600 women (mean [SD] age, 30.7 [4.3] years) were enrolled and randomized; all were included in the intention-to-treat analysis. Immediately after fetal delivery, the score on the numeric rating scale of pain was lower with esketamine (median [IQR], 0 [0-1]) than with placebo (median [IQR], 0 [0-2]; median difference, 0; 95% CI, 0-0; P = .001), but the difference was not clinically important. The Ramsay Sedation Scale scores were higher (sedation deeper) with esketamine (median [IQR], 4 [3-4]) than with placebo (median [IQR], 2 [2-2]; median difference, 2; 95% CI, 2-2; P < .001). The neonatal Apgar scores did not differ between the 2 groups at 1 minute (median difference, 0; 95% CI, 0-0; P = .98) and at 5 minutes (median difference, 0; 95% CI, 0-0; P = .27). Transient neurologic or mental symptoms were more common in patients given esketamine (97.7% [293 of 300]) than in those given placebo (4.7% [14 of 300]; P < .001). Conclusions and Relevance: For women undergoing cesarean delivery under epidural anesthesia, a subanesthetic dose of esketamine administered before incision produced transient analgesia and sedation but did not induce significant neonatal depression. Mental symptoms and nystagmus were common but transient. Indications and the optimal dose of esketamine in this patient population need further clarification, but study should be limited to those who require supplemental analgesia. To improve prognosis of cancer patients and determine the integrative value for analysis of disease-free survival prediction, a clinic investigation was performed involving with 146 non-small cell lung cancer (NSCLC) patients (83 men and 73 women; mean age: 60.24 years ± 8.637) with a history of surgery. Their computed tomography (CT) radiomics, clinical records, and tumor immune features were firstly obtained and analyzed in this study. Histology and immunohistochemistry were also performed to establish a multimodal nomogram through the fitting model and cross-validation. Finally, Z test and decision curve analysis (DCA) were performed to evaluate and compare the accuracy and difference of each model. In all, seven radiomics features were selected to construct the radiomics score model. The clinicopathological and immunological factors model, including T stage, N stage, microvascular invasion, smoking quantity, family history of cancer, and immunophenotyping. The C-index of the comprehensive nomogram model on the training set and test set was 0.8766 and 0.8426 respectively, which was better than that of the clinicopathological-radiomics model (Z test, P =0.041<0.05), radiomics model and clinicopathological model (Z test, P =0.013<0.05 and P =0.0097<0.05). Integrative nomogram based on computed tomography radiomics, clinical and immunophenotyping can be served as effective imaging biomarker to predict DFS of hepatocellular carcinoma after surgical resection.

Background: Necroptosis is a form of programmed cell death, and studies have shown that long non-coding RNA molecules (lncRNAs) can regulate the process of necroptosis in various cancers. We sought to screen lncRNAs associated with necroptosis to predict prognosis and tumor immune infiltration status in patients with hepatocellular carcinoma (HCC). Methods: Transcriptomic data from HCC tumor samples and normal tissues were extracted from The Cancer Genome Atlas database. Necroptosis-associated lncRNAs were obtained by co-expression analysis. Necroptosis-associated lncRNAs were then screened by Cox regression and least absolute shrinkage and selection operator methods to construct a risk model for HCC. The models were also validated and evaluated by Kaplan-Meier analysis, univariate and multivariate Cox regression, and time-dependent receiver operating characteristic (ROC) curves. In addition, Gene Ontology, Kyoto Encyclopedia of Genes and Genomes enrichment, gene set enrichment, principal component, immune correlation, and drug sensitivity analyses were applied to assess model risk groups. To further differentiate the immune microenvironment of different HCC subtypes, the entire dataset was divided into three clusters, based on necroptosis-associated lncRNAs, and a series of analyses performed. Results: We constructed a model comprising four necroptosis-associated lncRNAs: POLH-AS1, DUXAP8, AC131009.1, and TMCC1-AS1. Overall survival (OS) duration was significantly longer in patients classified as low-risk than those who were high-risk, according to our model. Univariate and multivariate Cox regression analyses further confirmed risk score stability. The analyzed models had area under the ROC curve values of 0.786, 0.713, and 0.639 for prediction of 1-, 3-, and 5-year OS, respectively, and risk score was significantly associated with immune cell infiltration and ESTIMATE score. In addition, differences between high and low-risk groups in predicted half-maximal inhibitory concentration values for some targeted and chemical drugs, providing a potential basis for selection of treatment approach. Finally, cluster analysis facilitated more refined differentiation of the immune microenvironment in patients with HCC and may allow prediction of the effectiveness of immune checkpoint inhibitors. Conclusions: This study contributes to understanding of the function of necroptosis-related lncRNAs in predicting the prognosis and immune infiltration status of HCC. The risk model constructed and cluster analysis provide a basis for predicting the prognosis of patients with HCC and to inform the selection of immunotherapeutic strategies.

In recent years, the application of low-refractive-index materials in the optical field has attracted considerable attention due to it high transmittance and high optical sensitivity. In this study, we synthesized SiO2 porous hollow spheres (SPHS) with an ultralow refractive index (n = 1.05) by using a templating method. Their refractive indices could be easily controlled from 1.05 to 1.08 by tuning the thickness of shell. In addition, a droplet coatings method is proposed for SPHS colloidal crystal (CC) by controlling the temperature and humidity. The SPHS CCs displayed distinct structural colors when the incident angle was adjusted and demonstrated high angular resolution. Moreover, the iridescent color changes could be observed with the naked eye. For surface-enhanced Raman scattering application, more analyte could be absorbed by the porous shells, and metal nanoparticles were coated on the SPHSs surface to increase the hot spot density for improving the SERS intensity.

OBJECTIVE: To explore the correlation between radiomic features and the pathology of pure ground-glass opacities (pGGOs), we established a radiomics model for predicting the pathological subtypes of minimally invasive adenocarcinoma (MIA) and precursor lesions. METHODS: CT images of 1521 patients with lung adenocarcinoma or precursor lesions appearing as pGGOs on CT in our hospital (The Third Affiliated Hospital of Sun Yat-sen University) from January 2015 to March 2021 were analyzed retrospectively and selected based on inclusion and exclusion criteria. pGGOs were divided into an atypical adenomatous hyperplasia (AAH)/adenocarcinoma in situ (AIS) group and an MIA group. Radiomic features were extracted from the original and preprocessed images of the region of interest. ANOVA and least absolute shrinkage and selection operator feature selection algorithm were used for feature selection. Logistic regression algorithm was used to construct radiomics prediction model. Receiver operating characteristic curves were used to evaluate the classification efficiency. RESULTS: 129 pGGOs were included. 2107 radiomic features were extracted from each region of interest. 18 radiomic features were eventually selected for model construction. The area under the curve of the radiomics model was 0.884 [95% confidence interval (CI), 0.818-0.949] in the training set and 0.872 (95% CI, 0.756-0.988) in the test set, with a sensitivity of 72.73%, specificity of 88.24% and accuracy of 79.47%. The decision curve indicated that the model had a high net benefit rate. CONCLUSION: The prediction model for pathological subtypes of MIA and precursor lesions in pGGOs demonstrated a high diagnostic accuracy. ADVANCES IN KNOWLEDGE: We focused on lesions appearing as pGGOs on CT and revealed the differences in radiomic features between MIA and precursor lesions. We constructed a radiomics prediction model and improved the diagnostic accuracy for the pathology of MIA and precursor lesions.

The development of the effective diagnostic method for the determination of cancer biomarkers is one of the most promising strategies for early clinical diagnosis of cancer. Here, based on the preparation of heterogeneous cuprous oxide coated silver (Ag@Cu2O) nanocomposites/graphitic carbon nitride (g-C3N4)/cadmium sulfide (CdS) nanoarrays structure, a highly sensitive photoelectrochemical (PEC) biosensor for the examination of carcinoembryonic antigen (CEA) has been constructed successfully. The combination of photoactive semiconductor materials g-C3N4 and CdS increases the electron transfer rate between them and enhances their photocurrent response, thus greatly increasing the concentration detection range. At the same time, the specific recognition between antigen and antibody is used to form a sandwich structure secondary antibody (Ab2)/CEA/antibody (Ab1). And because Ag@Cu2O has the function of absorbing light and consuming electron donor. Therefore, the successful measurement of CEA was achieved by labeling Ag@Cu2O on Ab2 and finally immobilizing it on the sensor to correlate the current reduction with the CEA concentration. The sandwich PEC biosensor proposed by this signal amplification strategy under optimal conditions has good analytical performance for CEA, with a wide linear detection range (from 10-5 to 1 ng/mL) and a low detection limit of 0.0011 pg/mL. The PEC biosensor constructed by this method showed high sensitivity, excellent anti-interference ability, favourable repeatability, and good stability.

Evidence has demonstrated that Daphnetin has antiangiogenesis activity, indicating it might be a new multi-targeted medication for cancer therapy. Here, we aimed to reveal Daphnetin role in hepatocellular carcinoma (HCC) progression and the underlying mechanism. Huh7 and SK-HEP-1, two human HCC cell lines were used in this study. MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide), colony formation, flow cytometry, and tumor-bearing experiments were applied to evaluate the effects of different concentrations of Daphnetin on cell viability, apoptosis, cell cycle, and in vivo tumor formation, respectively. Real-time PCR (Polymerase Chain Reaction)and western blotting were applied to measure the mRNA and protein levels of ß-catenin. We observed that Daphnetin inhibited cell viability and tumorigenesis, promoted cell apoptosis, and induced a G1 phase arrest in a dose-dependent manner in both Huh7 and SK-HEP-1 cells, which were rescued by SKL2001, an activator of the Wnt/ß-catenin signaling. Taken together, this study reveals that Daphnetin exerts an antitumor role in HCC through the inactivation of Wnt/ß-catenin signaling.

A surge of effort has been devoted to establishing super-wetting membranes with versatility for oily waste water purification. However, persistent challenge remains the lower separation flux. Moreover, the majorities of catalysts are only adsorbed on the surface and easily fall off after multiple cyclic separations. In this work, an effective strategy has been taken to construct a composite membrane consisting of polyacrylic acid functionalized carbon nanotubes (CNTs-PAA) and MIL101(Fe)@platinum nanoparticles (MIL101(Fe)@Pt NPs). The obtained CNTs-PAA/MIL101(Fe)@Pt composite membrane can achieve degradation of dye molecules and at the same time effective separation of oil-in-water emulsions. The separation throughput of this composite membrane can reach up to 11000 L m-2 h-1 bar-1, which has exceeded most of the previous reported multifunctional separation membranes. Furthermore, this composite membrane has presented stable mechanical property and excellent anti-corrosion ability. This work gives comprehensive consideration to excellent separation performance, versatility and stability, which could have potential applications in practical oily wastewater treatment.

Upconversion luminescence is an optical process that emits high-energy photons by absorbing two or more low-energy photons, which is caused by anti-Stokes luminescence, has attracted significant attention in recent years. Upconversion nanomaterials are superior to quantum dots and organic dyes because of their unique properties, including non-photobleaching, non-blinking, low-toxicity, photostability, high conversion efficiency, high light penetration in biological tissues and long lifetime, from nanoseconds up to 6 orders of magnitude to milliseconds, which render them an ideal kind of fluorescent labels for a variety of analytical formats as well as bioimaging in cancer therapy, showing great potential for both in vitro and vivo imaging and biodetection assays applications. This paper will introduce upconversion luminescence nanomaterials from mechanisms composition and synthesis, moreover, we will also discuss recent progress regarding the applications of upconversion nanomaterials in sensing, imaging and therapy.

As an ideal alternative to bisphenol A (BPA), bisphenol S (BPS) has similar biotoxicity, teratogenicity, carcinogenicity and mutagenicity as BPA. Nevertheless, to date, a fast, sensitive and portable method for meeting on-site measurement of BPS had not been established. Hence, it was particularly urged to develop a fast and highly sensitive method for tracing BPS. Currently, a novel molecularly imprinted polymer (MIP) electrochemical sensor had been successfully fabricated for the detection of BPS, wherein the composite of three-doped carbon quantum dots (B, N, F-CQDs) and silver nanoparticles (AgNPs) were utilized as an electron conducting layer, and MIP was applied as recognition element of target molecules. The step-by-step fabrication process and the adsorption capacity of the modified electrode were evaluated by cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS) and chronocoulometry (CC). The results indicated that the synergy between B, N, F-CQDs and AgNPs dramatically improved the sensitivity of the electrode and achieved the amplification of the electrical signal. Meanwhile, the electrochemical activities of BPS were explored by CV and differential pulse voltammetry (DPV). And, the various parameters relating to the electrochemical kinetic properties of BPS were calculated. To the best of our knowledge, this was rarely reported in peer journals. The MIP remarkably improved the selectivity of the sensor owing to the specific recognition of the imprinted cavities. The linear response range of the new sensor was 1 × 10-8 M to 5 × 10-5 M with a detection limit of 1.12 × 10-8 M and the electrode designed in this paper could meet the requirement of trace-level measurement of BPS in biological and environmental samples. Additionally, the sensor was used to determine BPS in plastic products with good anti-interference and acceptable recovery.

Tissue-enriched genes are highly expressed in one particular tissue type and represent distinct physiological processes. The dynamic profile of tissue-enriched genes during tumorigenesis and progression remains largely unstudied. Here, we identified tissue-enriched genes from 12 tissue types based on RNA sequencing data from the Cancer Genome Atlas (TCGA), and found that the liver had the largest number of such genes among the 12 tissue types. The characteristics of liver-enriched genes were further investigated. Most liver-enriched genes were downregulated and metabolism-related genes, which were associated with pathological stage and dedifferentiation in patients with hepatocellular carcinoma (HCC). Hypermethylation might be a mechanism underlying the downregulation of liver-enriched genes. We constructed a liver-enriched gene set and demonstrated that it is associated with the prognosis of the patients with HCC both in the TCGA cohort and the Gene Expression Omnibus (GEO) datasets. Moreover, we discovered that the degree of the dissimilarity between tumors and normal tissues was correlated with the prognosis of patients with HCC and the biological behaviours of the tumors. These results will help identify prognostic biomarkers of patients with HCC, and enhance our understanding of the molecular mechanisms of hepatocarcinogenesis and progression.

OBJECTIVE: To investigate the efficacy of Chaihu Ji Sang Decoction (CJSD), (a modification of Xiao Chaihu Tang (Minor Decoction of Bupleurum) was evaluated in patients with post-infection cough (PIC). Its effect on cough reflex sensitivity (CRS) was also described. METHODS: One hundred and eighteen patients with PIC were enrolled and randomly divided into an experimental group and control group (54 patients in each group). The experimental group was given CJSD orally, and the control group was given cetirizine hydrochloride for 7 d. Cough symptom scores and TCM symptom scores were compared before and after treatment. Efficacy, cough onset time, recurrence, and adverse drug reactions were additionally evaluated. Finally, CRS was evaluated before and after treatment of PIC patients in the experimental group. RESULTS: Fifty-two patients in the experimental group and fifty control patients completed the study. Cough symptom scores and the TCM symptom scores of the two groups were significantly improved after treatment, however cough symptoms in the experimental group 1 (0, 2) were significantly lower than the control group 2 (1, 2) after treatment (P < 0.05). Similarly, TCM symptom scores 4 (0, 7) in the experimental group were significantly lower than the control group 6 (3, 12) (P < 0.01). Overall, the effective rate of the experimental treatment (84.60%) was significantly higher than that of the control treatment (68.00%) (P < 0.01). The onset time of efficacy of the experimental treatment was one day faster than the control treatment [2 (2,3) d vs 3 (2,3) d, P < 0.01]. The relapse rate in the experimental group (9.09%) was lower than that of the control group (29.41%) (P < 0.05). 39 PIC patients in the experimental group completed a capsaicin cough challenge test before treatment. The lgC2 and lgC5 of the experimental group prior to treatment was 1.19 (0.59, 1.49) and 1.80 (1.49, 2.10), respectively. The lgC2, lgC5 of the healthy control group was 1.49 (0.89, 2.40) and 2.70 (2.40, 3.00), respectively. The differences in lgC2 and lgC5 between experimental group and healthy groups were significant at this time point (both P < 0.01). After treatment, 34 patients in the experimental group received re-examination. lgC5 increased from 1.80 (1.49, 2.10) to 2.10 (1.49, 3.00). CRS after treatment/re-examination was significantly lower than those of baseline in the experimental group (P < 0.05). CONCLUSION: CJSD could effectively improve the clinical symptoms of PIC patients, with demonstrable low recurrence and safety. CJSD also significantly decreased the CRS in patients with PIC.

RESUMO

OBJECTIVE: To evaluate the efficacy of daily low-dose tadalafil therapy for overactive bladder (OAB) in women. PATIENTS AND METHODS: A total of 96 women with idiopathic OAB from 3 medical centers in the south of Zhejiang Province of China were randomly assigned to treatment with daily low-dose tadalafil (5 mg, n = 48) or placebo (n = 48) for 3 months. The Indevus Urgency Severity Scale, overactive bladder symptom score (Homma et al, 2006), and a 3-day micturition diary with frequency, incontinence, and urgency episodes were recorded and compared before the treatment, every 2 weeks following the treatment, and 3 months after the treatment. Uroflowmetry and transabdominal ultrasound were also conducted following the treatment to determine the maximum flow rate, voided volume, postvoid residual volume, total bladder capacity, and voiding efficiency. The patient's overall rating of improvement in symptoms was assessed as well. RESULTS: The overactive bladder symptom score significantly decreased, and the frequency, incontinence, and urgency episodes significantly improved in the tadalafil treatment group as compared with the placebo group and baselines at weeks 4, 6, 8, 10, and 12, as well as 3 months posttreatment (P <.05). In addition, voided volume and total bladder capacity obviously increased in the treatment group (P <.05). The Indevus Urgency Severity Scale decreased from week 4 to 3 months posttreatment in the treatment group (P <.05). No changes were found in the maximum flow rate, postvoid residual volume, and voiding efficiency. All adverse symptoms were mild to moderate. CONCLUSION: Daily low-dose tadalafil is a considerable, well-tolerated, and effective treatment for OAB in women.

A yellow-leaf mutant (yl11) with chlorophyll-deficient in rice (Oryza sativa L.) was selected from the progeny of a japonica rice variety "Jiahua 1" treated with 60Co γ-radiation. In comparison with the wild-type parent, "Jiahua 1", the mutant had yellow-leaves at whole growth stages and displayed significantly decreased in chlorophyll content and net photosynthetic rate. Underdeveloped chloroplast and alterations of the major agronomic traits, such as plant-heights, were also observed in the mutant. Genetic analysis confirmed that the yellow-leaf mutant trait was controlled by a single recessive nuclear gene (yl11). Using SSR and In/Del molecular markers and 920 F2 and F3 plants from the cross of yl11 with the indica variety Peiai 64S, the yl11 was mapped between the molecular markers MM2199 and InDel21039 with a physical distance of 110 kb on the long arm of chromosome 11, in which no known functional genes for chlorophyll synthesis or chloroplast development in rice has been found. These findings will provide a foundation for the cloning and functional analysis of this gene in the future.

OBJECTIVE: To study the emotional and depressive differences between severe acute respiratory syndrome (SARS) patients whose occupations were doctor/nurse and others. METHODS: During the three months when SARS was explosive in 2003, 524 questionnaires were collected from Xuanwu Hospital, You'an Hospital, Xiaotangshan Hospital, Renmin Hospital and Ditan Hospital in Beijing. There were 109 questionnaires finished by patients as doctors/nurses themselves. For a background matching, 109 questionnaires were also finished by the others. RESULTS: From 218 questionnaires, we found that the score on emotional condition (46.6204 +/- 8.4408 vs. 41.6789 +/- 8.612 95, P < 0.001) of SARS patients whose jobs were doctor/nurse was higher than the other groups on while the score of SARS patients whose jobs were doctor/nurse was lower than the other groups (37.7615 +/- 9.026 61 vs. 41.2844 +/- 9.655 25, P = 0.006) regarding depressive condition. The scores of emotional and depressive condition were all correlated with the factor as "I can not master my future". CONCLUSION: Doctors/nurses having SARS had less emotional and depressive conditions than the others, which might due to the difference in medical knowledge, working condition and the route of infection, suggesting that psychological intervention in the post-SARS period called for attention.
